Prostaglandin active pharmaceutical ingredients (APIs) require specific expertise and experience to synthesize, purify, and handle. Cedarburg Hauser Pharmaceuticals has placed a special emphasis on the development of a new, more efficient synthesis for purifying and handling prostaglandins. Cedarburg Hauser Pharmaceuticals was contracted for the development and validation of a new synthetic process for a commercially available prostaglandin analog. The existing synthetic process was tedious, and required multiple steps, isolations and purifications. In the development of this compound, we successfully utilized our experience with delta-9-THC, also a potent, sticky oil, as well as our expertise with chromatographic techniques to safely and efficiently manufacture this important prostaglandin analog. In addition to reducing the overall number of synthetic steps and introducing efficiencies that made the synthesis significantly more cost-effective, a thorough understanding of the process enabled the implementation of process controls that simplified purification. Modifying the workups enabled the removal of two of the three chromatography steps, reducing the number to a single high yielding normal phase chromatographic process and producing material that exceeded our customer's target specifications.
